S07-03 Asymmetric division in zebrafish neural tube is a non-stochastic event and the neuron results from the daughter that inherits the apical domain

چکیده
Emerging evidence suggests that neurogenesis may occur within the adult hypothalamus but the character and location of hypothalamic neural stem cells is unknown. We have identified a stem cell niche, confined to the central/posterior hypothalamus. Niche-derived cells require FGF signalling to maintain their proliferation exvivo, and proliferate, but do not differentiate, in response to exogenous FGF2 invivo. Enhanced proliferation of hypothalamic neural stem cells occurs concommitant with a reduction in proliferating progenitors. We define two distinct FGF2-dependent cell types, whose position correlates with particular tanycyte populations. Cells that map to GFAP+ive tanycytes show hallmarks of classical neural stem cells; cells that map to adjacent GFAPive ventral a2 tanycytes show characteristics intermediate beween stem and progenitor cells. Both cell types appear largely quiescent invivo, but endogenous FGFs may govern their proliferation: FGF ligands are expressed specifically within different tanycyte populations and FGF signalling elements are stochastically detected in tanycyte cells.

Intralineage Directional Notch Signaling Regulates Self-Renewal and Differentiation of Asymmetrically Dividing Radial Glia
Asymmetric division of progenitor/stem cells generates both self-renewing and differentiating progeny and is fundamental to development and regeneration. How this process is regulated in the vertebrate brain remains incompletely understood. Here, we use time-lapse imaging to track radial glia progenitor behavior in the developing zebrafish brain. We find that asymmetric division invariably gene...
متن کاملMitotic spindle orientation can direct cell fate and bias Notch activity in chick neural tube
Inheritance of apical membrane is proposed to maintain vertebrate neural stem cell proliferation. However, evidence for this is contradictory. Using direct clonal analysis and live imaging in chick neural tube, we show that divisions that separate apical and basal components generate an apical daughter, which becomes a neuron, and a basal daughter, which rapidly re-establishes apico-basal polar...
متن کاملCyclinD2 at the edge: splitting up cell fate.
Asymmetric cell division and cell cycle length are two fundamental mechanisms that influence the fate of neural stem cells during mammalian brain development. In this issue of The EMBO Journal, the team of Noriko Osumi proposes an elegant link between the two by showing that the mRNA of cyclinD2, a positive regulator of G1 progression, is confined to the basal end-foot of radial glial cells and...
متن کاملThe neural crest epithelial-mesenchymal transition in 4D: a 'tail' of multiple non-obligatory cellular mechanisms.
An epithelial-mesenchymal transition (EMT) is the process whereby epithelial cells become mesenchymal cells, and is typified by the generation of neural crest cells from the neuroepithelium of the dorsal neural tube. To investigate the neural crest EMT, we performed live cell confocal time-lapse imaging to determine the sequence of cellular events and the role of cell division in the EMT. It wa...
متن کاملMonitoring neural progenitor fate through multiple rounds of division in an intact vertebrate brain.
The behaviour of neural progenitors in the intact vertebrate brain and spinal cord is poorly understood, chiefly because of the inaccessibility and poor optical qualities inherent in many model systems. To overcome these problems we have studied the optically superior brain of the zebrafish embryo and have monitored the in vivo behaviour of fluorescently labelled neural progenitors and their da...
